Homeif i use buff Intelligence spell and sustain it, will that spell mana cost get reduction from after the intelligence get buffed or before that spell activated???

Spellhowler and Spellsinger let you affect Intelligence. A staff and other effects that explicitly state so can debuff Intelligence too.
If Intelligence is debuffed, the mana reduction gets debuffed presumably after that effect takes place.
